Six hours is just enough time to do the trip from Seattle around Mount Rainier National Park. Go in the Nisqually entrance (SW corner of the park) go up to Paradise, then down through Stevens Canyon to Ohanapacosh, then north over Cayuse Pass and if you have the time a side trip up to Sunrise (Sunrise is a less popular destination than paradise, but well worth it), then back to hwy 410 and exit the NE corner of the park following hwy 410 back to the lowlands.
If the weather is too cloudy for Rainier and you can devote an entire (long day) or better overnight, head north of Seattle on I-5, then east on Hwy 20 over the North Cascade Hwy, then south to Wenatchee, then back west over Hwy 2 over Stevens Pass. This is the classic "cascades loup" trip and will give you a lot to see and you'll travel through many different types of ecosystems. It's one of my favorite road trips and you will almost always have good weather during a large part of the trip. Make a two day trip out of it if at all possible (I think it's nearly 500 miles RT).

I was going to recommend the North Cascade/ Hwy 20 loop to Winthrop. If you want to do some trout fishing while you're here, I'd look into the Lahontan Cutthroat trout around the Soap Lake area just a bit SE of Winthrop/Chelan. https://northwest-fly-fishing.myshopify.com/blogs/departments/76295429-omak-lake-waI've caught them in Lenore and they were almost as long as my arm. Sounds like Omak has some bruisers.

I would head out on hwy 20 into the North Cascades and drive for half the time you have available, then turn around and come back. Gorgeous country. Make sure you have a tank full of gas.
Depending on where you're at in the Seattle area, you might be able to get all the way to Winthrop and have an ice cream cone before heading back. It can take 3 hours just to get across seattle at the wrong time though.
